Taxane-Based Chemotherapy Is Effective in Metastatic Appendiceal Adenocarcinoma
Oncologist. 2023 Sep 20:oyad263. doi: 10.1093/oncolo/oyad263. Online ahead of print.ABSTRACTAppendiceal cancer is a rare, orphan disease with no therapies currently approved by the FDA for its treatment. Given the limited data regarding drug efficacy, these tumors have historically been treated with chemotherapy designed for colon cancer. However, an overwhelming body of molecular data has demonstrated that appendiceal adenocarcinoma is a distinct entity with key molecular differences from colon cancer, notably rare APC mutation. Recognizing that APC loss-of-function is thought to contribute to taxane resistance and that t...

Chemo-immune cell therapy by intratumoral injection of adoptive NK cells with capecitabine in gastric cancer xenograft model
CONCLUSION: Although NK cell therapy could effectively decrease the mitotic count in vivo, the obtained findings indicated lesser potency than MC despite ex vivo activation. In order to enhance NK cell therapy effectiveness, suppressive features of the tumor microenvironment and inhibitory immune checkpoints blockade should be considered.PMID:37736341 | PMC:PMC10509737 | DOI:10.34172/bi.2022.26386

Galectin-3 Cooperates with CD47 to Suppress Phagocytosis and T cell Immunity in Gastric Cancer Peritoneal Metastases
Cancer Res. 2023 Sep 22. doi: 10.1158/0008-5472.CAN-23-0783. Online ahead of print.ABSTRACTThe peritoneal cavity is a common site of gastric adenocarcinoma (GAC) metastasis. Peritoneal carcinomatosis (PC) is resistant to current therapies and confer poor prognosis, highlighting the need to identify new therapeutic targets. CD47 conveys a "don't eat me" signal to myeloid cells upon binding its receptor SIRPα, which helps tumor cells circumvent macrophage phagocytosis and evade innate immune responses. Previous studies demonstrated that the blockade of CD47 alone results in limited clinical benefits, suggesting that other t...

Population effectiveness of endoscopy screening for mortality reduction in gastric cancer
CONCLUSIONS: In addition to the effectiveness of the used modality, the disease burden and screened rate were important in the population effectiveness of cancer screening. Regarding the high NNSs and the low expected mortality-rate reduction, population-based endoscopic screening seems not to be effective in many countries, and these results are meaningful in decision-making regarding the introduction of endoscopic screening.PMID:37731836 | PMC:PMC10508325 | DOI:10.1002/deo2.296

Lifestyle habits and gastric cancer in an East Asian population: a Mendelian randomization study
CONCLUSIONS: Our study suggests that tea intake may reduce the risk of GC, for which smoking is a potential risk factor. Nevertheless, a larger and more diverse sample size is needed for further validation.PMID:37731647 | PMC:PMC10507616 | DOI:10.3389/fonc.2023.1224753

Serum basic fibroblast growth factor and interleukin-1 β predict the effect of first-line chemotherapy in patients with advanced gastric cancer
CONCLUSION: The FLOT regimen was effective in reducing the serum CEA, CA199, and TPS levels as well as in improving the 1-year survival rate of patients with good tolerability, making it worthy of clinical promotion and application.PMID:37731570 | PMC:PMC10507556 | DOI:10.12998/wjcc.v11.i26.6083

Exosomal circKIAA1797 Regulates Cell Progression and Glycolysis by Targeting miR-4429/PBX3 Pathway in Gastric Cancer
Biochem Genet. 2023 Sep 20. doi: 10.1007/s10528-023-10529-z. Online ahead of print.ABSTRACTIn recent years, circular RNAs (circRNAs) are extensively studied in the progression of various types of cancer, while the mechanism of circKIAA1797 is rarely studied in gastric cancer (GC). Hence, this research aimed to investigate the expression of exosomal circKIAA1797 and its biological function in GC cells. Exosomes were extracted from the serum of GC patients and identified by transmission electron microscopy (TEM) and nanoparticle tracking analyzer (NTA). CD81, CD63, Bcl-2, Bax, and pre-leukemia transcription factor 3 (PBX3) p...

Multidimensional Analysis of a Cell-Free DNA Whole Methylome Sequencing Assay for Early Detection of Gastric Cancer: Protocol for an Observational Case-Control Study
CONCLUSIONS: This is the first registered case-control study designed to investigate a stacked ensemble model integrating several cfDNA features generated from a bisulfite-free whole methylome sequencing assay. These features include methylation patterns, fragmentation profiles, and chromosomal copy number changes, with the aim of identifying the GC population. This study will determine whether multidimensional analysis of cfDNA will prove to be an effective strategy for distinguishing patients with GC from nonmalignant individuals within the Chinese population. We anticipate the THEMIS model will complement the standard-o...
